QUANTITATIVE COMPOSITION
Each tablet contains:
Ampicillin (trihydrate) ………1 gr.
Excipients: Magnesium stearate, microcrystalline cellulose.

PROPERTIES
Ampicillin is a semi-synthetic penicillin derived from the nucleus 6-amino-penicillanic acid. It has bactericidal action against susceptible organisms during the stage of active multiplication, acting through the inhibition of biosynthesis of cell wall mucopeptide.
It is acid stable. Absorption of ampicillin is 40% of the dose administered orally on an empty stomach oral %; the presence of food in the stomach will delay absorption. At the usual dose it reaches therapeutic concentrations in serum and various body fluids based on its use in the indications mentioned.
Ampicillin penetrates into the cerebrospinal fluid when the meninges are inflamed. It is excreted in the urine and bile at high concentrations and in biologically active form. 20% is bound to plasma proteins.
Ampicillin possesses broad-spectrum antibacterial activity against both gram-positive and gram-negative bacteria. It has shown to be effective in vitro against the following microorganisms, amongst others:
Gram-positive: Streptococcus species (including Streptococcus pneumoniae) and Non-penicillinase-producing staphylococci
Gram-negative: Haemophilus influenzae, Proteus mirabilis, Salmonella species,
Shigella species, Escherichia coli, Neisseria gonorrhoea and Neisseria meningitides.

INDICATIONS
It is indicated for the treatment of infections caused by susceptible strains of the abovementioned microorganisms. These include: ear, nose and throat (ENT) infections, respiratory tract infections, gastrointestinal infections and genito-urinary tract infections.

POSOLOGY
Usual dosage:
Adults: 1 tablet every 6 hours.
In severe infections this dosage may be increased, only if decided by your doctor. Ampicillin should be taken on an empty stomach half to one hour before meals. Treatment should be continued for a minimum of 48 to 72 hours beyond the time that the patient becomes asymptomatic
In the presence of renal impairment the dosage should be adjusted depending on the degree of impairment.
It is recommended that there be at least 10 days treatment for any infection caused by Beta-haemolytic streptococci.

CONTRAINDICATIONS
Britapen is contraindicated in patients known to be allergic to penicillin and should not be given to patients with infectious mononucleosis.

PRECAUTIONS
Caution should be exercised in patients with allergy to cephalosporins and in patients with a previous history of allergy mainly of medicamentosus origin.

INTERACTIONS
Concurrent administration of allopurinol during treatment with ampicillin can increase the likelihood of allergic skin reactions. Bacteriostatic drugs may interfere with the bactericidal action of ampicillin therefore its concomitant administration is not recommended.

SIDE EFFECTS
Allergic reactions, urticaria and erythematous skin reactions have been reported. The incidence of these side effects is higher in patients suffering from infectious mononucleosis.
Such reactions as leucopenia, neutropenia and eosinophilia have been reported during therapy. Moderate increase in transaminases has been reported, especially in children. These reactions are usually reversible on discontinuation of therapy.
Gastro-intestinal symptoms, nausea, vomiting and diarrhoea may occur.
If supra-infections with non-susceptible organisms occur therapy should be discontinued and appropriate therapy be instituted.

OVERDOSE AND ITS TREATMENT
Should an allergic reaction occur, antibiotic treatment should be discontinued and appropriate therapy be instituted.
In the case of an overdose or accidental ingestion, contact your local poison control centre.
